Giemsa Stain is a versatile and widely used differential stain designed for the visualization of blood cells, blood-borne parasites, and chromosomal structures. It is most commonly applied in hematology for staining peripheral blood and bone marrow smears, enabling detailed examination of white blood cells, red blood cells, and platelets.

Giemsa is also a gold standard stain in parasitology—essential for the detection of Plasmodium spp. (malaria), Trypanosomes, and other protozoan parasites in blood samples. In cytogenetics, it is used in G-banding for the karyotyping of chromosomes, providing sharp, reproducible banding patterns.

The stain contains azure, eosin, and methylene blue, which selectively bind to cellular components, producing blue to purple nuclei, pale cytoplasm, and pink to salmon-colored red blood cells. Giemsa can be used as a standalone stain or in combination with other protocols such as Wright-Giemsa for enhanced contrast and diagnostic clarity.
